Proteinase K (also known as protease K or endopeptidase K) is a broad-spectrum serine protease widely used in molecular biology. Proteinase K can digest native keratin (hair), hence, the name “Proteinase K”. It is commonly used because of its broad specificity, which makes it useful to clean nucleic acid complexes and to lyse cells. It has been used for the isolation of mRNA, and high-molecular-weight DNA and to inactivate other enzymatic activities. The enzyme was discovered in 1974 in extracts of the fungus Engyodontium album (formerly Tritirachium album). Proteinase K is commonly used in molecular biology to digest proteins and remove contamination from preparations of nucleic acid. The addition of proteinase K to nucleic acid preparations rapidly inactivates nucleases that might otherwise degrade the DNA or RNA during purification, hence ideal for these applications. Proteinase K is a cation. It is highly suited to this application since the enzyme is active in the presence of chemicals that denature proteins, such as sodium dodecyl sulphate and urea, chelating agents such as EDTA, sulfhydryl reagents, as well as trypsin or chymotrypsin inhibitors. Proteinase K is used for the destruction of proteins in cell lysates (tissue, cell culture cells) and the release of nucleic acids.
Market Dynamics
In order to accelerate and broaden the development program and to further diversify the company’s portfolio, the key market players are focusing on the adoption of growth strategies such as applications of the proteinase K enzyme in novel diagnostic kits which is attributed to the growth of the global Proteinase K market. For instance, according to an article published in the Journal Med, on December 25, 2020, Proteinase K has the potential to work as an affordable, fast, and easy alternative to nucleic acid extraction with only a minor loss of sensitivity. Hence, it was used in SalivaDirect, an alternative diagnostic procedure for COVID-19 that uses saliva instead of nasopharyngeal swabs.
