Isothermal nucleic acid amplification technologies have applications in the field of recombinant DNA and molecular biology techniques. These techniques are used in the detection and analysis of small quantity of nucleic acids. Polymerase chain reaction is based on isothermal nucleic acid amplification technology. The polymerase chain reaction (PCR) is most widely used method for DNA amplification. Polymerase chain reaction (PCR) is used for the detection and identification of infectious diseases, genetic disorders, and for research purposes. PCR consists of thermocycling machine that facilitates separation and amplification of DNA strands. Isothermal amplification of nucleic acids is a simple process that rapidly and efficiently accumulates nucleic acid sequences at constant temperature. Various isothermal amplification techniques have been developed as alternatives to polymerase chain reaction (PCR).

Isothermal amplification chemistry has also been used to diagnostics with great success and is utilized in several commercial molecular diagnostic platforms, serving large testing centers and point-of-care markets. The advantage of these methods is that the nucleic acids amplification can be carried out at constant temperature, unlike PCR that requires cyclic temperature changes. Moreover, isothermal amplification can be conducted directly in living cells. The thermodynamics process in which the temperature of the whole system remains constant over a period of time is called isothermal process. Thus, the constant temperature of any system makes the process an isothermal process. For example, melting of ice at zero degree is an example of an isothermal process.
