G-protein coupled receptors (GPCRs) constitute a prominent and diverse family of cell surface receptors that play pivotal roles in cellular signaling and communication. These receptors are integral membrane proteins characterized by their distinct structural features, including seven transmembrane helices, an extracellular N-terminus, and an intracellular C-terminus. The seven transmembrane domains create a characteristic structure often likened to a bundle of alpha helices, facilitating the receptor's insertion into the cell membrane. The activation of GPCRs is triggered by the binding of specific ligands, which can include hormones, neurotransmitters, and other signaling molecules. Ligand binding induces conformational changes in the receptor, leading to its activation. The ligand-binding site is typically located at the extracellular N-terminus of the receptor, and the binding event initiates a cascade of intracellular signaling events. One of the hallmark features of GPCRs is their interaction with G-proteins, which are heterotrimeric complexes consisting of alpha, beta, and gamma subunits. Upon activation of the GPCR, the receptor undergoes conformational changes that allow it to catalyze the exchange of GDP for GTP on the alpha subunit of the G-protein. This activation of the G-protein triggers downstream signaling pathways, influencing various cellular responses.
