A nucleic acid sample preparation is a process in which RNA and DNA is extracted and purified from biological samples that may contain tissue, blood, or other organic matter and prepare them for use. For different research processes such as clinical and medical research, life science or pharmaceutical research, nucleic acid sample preparation is a required step. The main two types of procedures of obtaining DNA or RNA from a sample are by phenol-chloroform extraction and column based extraction. In phenol- chloroform extraction, the DNA or RNA is isolated in aqueous phase while rest of sample is partitioned into the organic phase. In case of column based extraction, the nucleic acid remains adhered to column and the organic material is washed away. Further steps are cutting fragments, labeling and amplifying them. Apart from these, procedures such as centrifugation, electrophoresis, and affinity separation can also be used for obtaining pure nucleic acid. This purified nucleic acid has applications in genetic engineering, molecular diagnosis, and forensics research related to life sciences.
